Method Description

The ADRA is proposed to address the molecular initiating event of the skin sensitisation AOP—namely, protein reactivity—by quantifying the reactivity of test chemicals towards model synthetic amino acid derivatives containing either lysine (NAL) or cysteine (NAC). The relative concentrations of NAC and NAL are measured by high-performance liquid chromatography (HPLC) with gradient elution and UV detection at 281 nm. Percent depletion values are then calculated for both NAC and NAL to discriminate between skin sensitisers and non-sensitisers.
