Method Description

The Neutral Red Release (NRR) assay is a quantitative colorimetric cytotoxicity test using cell lines to measure the immediate toxic effect of a test substance on the cell membrane, resulting in a leakage of intracellular contents. If cells in culture are preloaded with vital dye (neutral red [NR]) and then exposed to a toxic substance, the amount of dye released from the cells will reveal the degree of membrane damage. The result of the test is expressed as the concentration of test substance that gives rise to the release of 50% of the NR from preloaded cells. Cell membrane damage is one of the principal events that lead to chemical-induced eye irritation, and it can be modelled using the NRR assay.
